چکیده

The isolation of adenosine tetraphosphate and pentaphosphate from samples of adenosine triphosphate has been reported by Marrian (1) and Sacks (2). The available evidence is consistent with a st,ructure in which the phosphate residues are linked by phosphoanhydride bonds to one another and are carried by the 5’-hgdroxyl group of the ribose moiety (l-4). During attempts to estimate guanosine triphosphate by ion exchange thin layer chromatography (5), it was found that the samples of commercial guanosine triphosphate investigated contained a component that migrated more slowly than guanosine triphosphate, and const, ituted between 5 and 107, of the absorbance at 252 rnp. This paper is concerned with the purification and analysis of this component, which has proved to be guanosine tetraphosphate.

Interferon- Gamma- Inducible Guanosine Triphosphate Cyclohydrolase 1 (GTP-CH1) Pathway Is Associated with Frailty in Egyptian Elderly

Background: Chronic low-grade inflammation may be a cardinal pathophysiologic feature in the pathogenesis of frailty. Interferon-gamma (INF-γ) is an understudied proinflammatory cytokine in frailty that induces many inflammatory pathways including the guanosine triphosphate cyclohydrolase 1 (GTP-CH1) pathway. Our aim was to evaluate the GTP-CH1 pathway in Egyptian frail elderly subjects. ...

Adenosine Triphosphate : Guanosine Monophosphate Phosphotransferase

Several laboratories have reported studies on individual enzymes that belong to the general class of adenosine triphosphate : nucleoside monophosphate phosphotransferases (EC 2.7.4.4), commonly termed nucleoside monophosphate kinases (l-7). Studies with partially purified enzyme preparations have indicated that a variety of nucleoside monophosphates can serve as alternative substrates for nucle...

Salmonella Reprograms Nucleotide Metabolism in Its Adaptation to Nitrosative Stress

The adaptations that protect pathogenic microorganisms against the cytotoxicity of nitric oxide (NO) engendered in the immune response are incompletely understood. We show here that salmonellae experiencing nitrosative stress suffer dramatic losses of the nucleoside triphosphates ATP, GTP, CTP, and UTP while simultaneously generating a massive burst of the alarmone nucleotide guanosine tetrapho...

The influence of bound GDP on the kinetics of guanine nucleotide binding to G proteins.

Purified guanine nucleotide-binding regulatory proteins, as either the oligomers or the isolated nucleotide-binding alpha subunits, display anomalous kinetics of nucleotide binding. This is due to the presence of tightly bound GDP in these preparations. The dissociation of bound GDP is the rate-limiting step for nucleotide binding. GDP can be removed by chromatography in the presence of 1 M (NH...

Interaction of guanosine nucleotides with elongation factor 2. II. Effect of ribosomes and magnesium ions on guanosine diphosphate and guanosine triphosphate binding to the enzyme.

The effects of ribosomes and Mg-2plus on the binding of GDP and GTP to elongation factor 2 (EF-2) have been studied by an improved filter-binding assay. Both ribosomes and Mg-2plus strongly inhibit the binding of GDP but have apparently no effect on the GTP binding to the enzyme. An apparent stimulation by ribosomes of GTP binding to EF-2 is time-dependent and parallels a concomitant increase o...
